Please I want to make sure about the polarization of THP-1 to Macrophages by using PMA, then the polarization of these macrophages to M2 by IL-4 or to M1 by LPS and INF gamma, does the medium that contains PMA, IL-4, LPS, IFN G, or any other cytokines must be free-serum and free-antibiotics or not?

For cultivation of THP-1 cells we use RPMI medium containing 10 % heat-inactivated fetal bovine serum (FBS), 1 mM sodium pyruvate, 2 mM L-glutamine, 1 % PenStrep and 100 μg/ml Normocin.
For differentiation of THP-1 cells into more macrophage-like cells, we remove the cultivation medium by centrifugation and resuspend the cells in a fresh medium containing 10 % heat-inactivated FBS and 100 nM PMA without further supplements and without antibiotics.
Short: Your differentiation medium does not have to be serum-free, but I would recommend not to use antibiotics in the differentiation medium.
