What i mean to ask is whether an increase in fluorescence of nonylacridine orange staining along with an increase in mitotracker red fluorescence may be attributed as Mitochondrial fission?
Of course, its known that p-DRp 1 levels would increase during fission, but can flowcytometric measurements can be used to conclude the above phenomenon as it is a quick method in comparison to immunoblotting.

no you can not claim from the increase of fluorescence signal that cells Show a higher mitochondrial fission process. The increase of your fluorescence signal with mitotracker and nonylacridine orange could be interpreted by a higher stability of the mitochondrial membrane potential. but this is just a little bit more complicated. An increase in CL per se does not tell you anything about the integritiy of your mitochondrial function. You should also look for the distribution of your CL species, which can directly affect diverse mitochondrial functions leading to an affected mitochondrial membrane potentaial. So you also should look for the integrity of your complex activities as well as the Membrane potential.
To adress the fission process you should look for electron microscopy.

Thank you for your suggestion. Yes, I do have checked for mitochondrial complex activity and permeabilization and potential. I think I would rather go for microscopy and western to address the fission issue.
