Hello!

I have a lot of practice with lentiviruses (shRNA, knock-in, CRISPR, etc.). And now I need to transduce human and mouse cells by retroviruses (knock-in). I recieved plasmid pQCXIP and in that vector I have cloned my gene of interest via MCS. Sequence of pQCXIP is available.

Also I recieved plasmid named "gag-pol", but I have know idea what is the sequence. What is it in general. I suppose that plasmid is like psPAX2 for lentiviruses.

From the Internet I know that to produce retroviruses I need VSV-G plasmid and gag-pol. For Lentiviruses I use pMD2.G and psPAX2, respectively, and always have high titer and good results. But LTR and psi sequenses in lentivirus and retroviruses are various.

I know that for production of retroviruses Phoenix cells (or like GR-2... I can forget the name) could be used. I haven`t got these cells, only HEK293T and HEK293FT cells.

Furthermore, I need a protocol of AAVs. And retroviruses. It is desirable that the protocols are detailed and intended for HEK293T or HEK293FT cells. And I like detailed protocols=)

Can anyone help me please?
