We have been concentrating HIV pseudovirus in the lab using PEG6000. Upon centrifuging the PEGylated supernatants, we resuspend the PEG pellet in a cell culture medium. We do not remove the PEG from our virus preps. My question is whether PEG would affect in anyway mononuclear cells that we infect with our pseudovirus. Would it be better to avoid PEG and use the traditional ultracentrifugation instead? Does anyone have experience with this?
