DYNAL beads are a great way to purify T-cells from bulk PBMC cultures. There are positive and negative selection strategies. I've heard it said that positive selection using DYNAL beads can induce unwanted activation. We often lyse the cells for molecular work after bead-sorting. The whole procedure only takes 30mins. I wouldn't have thought non specific activation is a problem in this short time. Do others have an opinion on this?
