According to some authors: Lipase activity index, is a qualitative test it compares the diameter of a colony and the diameter of halo of lipase activity:
Lipase activity index (LAI), a rate between the halo diameter close to the colony
and the diameter of the colony during 15 days, with five replicates. The activity can be measured at different temperatures (30◦, 35◦, 40◦, 45◦C, etc.). Optimal temperature or better physiological temperature is the choice.
But It is better to use a quantitative test.
Isolate your strain
Do cultivation in optimal media for lipase production
After growth (early stationary phase) recover supernatant by centrifugation (10000 x, 5 min)
Measure lipase activity, in the supernatant, by quantitative test, for example using p-nitrophenyl miristate method (see attached article).
