we are trying to infect mice (BALB/c) with HSV-1F. but no visible changes are there on the skin like mentioned in literature (blisters on the area of infection). We administered intradermally and also by scratching the skin and titer given at 10^6 PFU. the virus used is being passaged in vero until titre 10^8 PFU. And virus used was freshly isolated.
please give possible suggestions regarding the issue.
hsv1 causes infection via contact with a skin of the infected person and hsv2 via sexual contact. so hsv1 or 2 doses not infected via blood on the other hand hsv1 or 2 transmitted via neuronal way inside body .hsv1 or2 are insert to latent state after infection. reactivation occur under specific conditions (immunity disorder state) that cause store more appear around a lip
I haven't seen an intradermal infection done with HSV in mice, but more likely scarification on the flank or cornea, footpad injection, intraperitoneal, intranasal, or intracranial injection. Did you find a good scarification protocol? Some of those are very specific as to the method of denuding the skin (Nair) and the number and types of scratches on the flank prior to virus administration. Also, 10 uL of your inoculum seems a little on the high side (if you have 10^8 PFU/ml and are giving 10^6 PFU), with 2 uL of a more concentrated virus being the norm.
What is the source of your virus? Minimal passage in Vero cells should be alright for maintaining its genotype, but you may want to either make sure it's Strain F or get a more virulent strain like McKrae.
That said, this figure shows that 10^5 PFU of HSV-1 F given by scarification gave 100% mortality 9 days postinfection. They don't seem to give an inoculation volume but their skin abrasion protocol is given.
