Usually lasts about a month before I smell that something has gone bad. I believe it is likely the FBS that spoils but wondered if old glycine could also cause the issue. Thanks
Glycine in solution, with yeast extract as a nutrient, can be degraded by to ammonia, carbon dioxide, and water by bacteria. A 10x stock solution is supposed to last more than 1 year but will oxidize and start turning yellow with time. Sigma-Aldrich adds preservatives to the 0.6 M solution that they sell and they recommend to store it between 2-8°C. Indeed it is much more likely that the FBS is to blame but if you suspect your glycine stock just make two block buffers, one with and one without glycine. Place them in the incubator for 1 - 2 days and you will have your answer. Either way, if your glycine stock is old you should just get another one. It's not expensive.
