Does anyone knows how to prepare zarrouk medium properly i found a recipe on int but the media becomes white after autoclave..please help ?

zarrouk medium precipitation..recipe etc.

You CANNOT autoclave Zarrouk medium because, at high temperature, phosphate will complex to calcium and magnesium, producing white precipitates.

At first, You have to prepare a sodium bicarbonate solution (16.8 g/L), then add some 5N NaOH to a final pH of 9. Filter at 0.22 micron and keep in refrigerator until use.

For other nutrients, I prepare stock solutions 100X or 1000X (depending on salt solubility) and autoclave separately.

Finally, I add nutrients stock solutions to filtered bicarbonate solution.
