Does anybody know how to isolate C Fragment from Tetanus toxoid (Not Toxin)? I am trying to use Papain to cleave the di-sulphide bonds, but I want to know what are the ideal conditions that need to be used. I am trying 16 hours at 25 degrees, and I am getting a fragment of about 40 kd. However, I think C fragment should be 50 kd. Will isolating the same from Toxoid and not toxin make a difference?

Which strain are you used whether that is Spore forming or non spore forming, You may be try with the Cross flow filtration (CFF) technology subsequently process to Affinity chromatography with Cellolose based ester ligand with gradient buffer elutent. As per my knowledge
