I currently have a trouble in size analysis of sulfo-Cy5 labelled nanoparticles by DLS possibly due to spectral property of the dye, of which excitation wavelength (646nm) overlaps with a laser of the machine (Malvern Nano Z with He-Ne laser 633nm). I suppose disrupting the Cy dye structure (by photobleaching or chemical oxidation?) could enable DLS analysis of the particles. Since the particles are both pH and reduction sensitive, mild conditions are required to disturb the dye. Does anyone know how to disrupt sulfo-Cy5 structure to change its spectral property?
