In one of our experiments we froze down mouse brain tissue with the intent of assessing specific parameters through histology. However, we would like to assess caspase activity in the tissue as well using the Caspase-Glo assay. According to the manufacturers this is possible with tissue, provided that the it was frozen and not fixed with formalin. I snap froze my samples in isopentane to prevent freeze fracture and now I am uncertain whether or not this may have affected the caspase activity. Could anyone perhaps give me some guidance please?
