when dissolving the FDA in acetone keep in mind to dilute the FDA stock approximately 1:1000. Otherwise acetone may damage the pollen grain membranes. Usually 2 mg FDA are dissolved per ml acetone. The stock solution can be stored at -20 °C for a few months. Add 1 µl FDA stock per ml of pollen medium and use the staining solution within half an hour to avoid precipitation of FDA.
However, the best test for pollen viability is to germinate them in germination buffer!
I usually prefer DMSO for FDA stock solutions because it can be stored in normal reaction tubes in the freezer. For acetone stocks I prefer small glass vessel with a screw lid. Actually, it doesn't matter which solvent you're using, I never observed difference in staining pattern.
