I would like to perform whole-mount in situ hybridization on fixed tissues that have been embedded in OCT compound and stored at -80°C.
Has anyone here done this procedure before? And if possible, are there any changes to the coloring procedure for whole-mount in situ hybridization?
The samples were fixed in 4% PFA, embedded in OCT after sucrose substitution following standard protocol, and are currently stored at -80°C.
I would be grateful for any help or advice.
