Hi, I have tried a designed protocol to purify IgEs from patient's serum but I haven't obtain the whole molecule (I do not know why). The protocol consists on incubating the serum on a CAP nitrocellulose membrane that is covered with anti-IgE antibody. The serum is then eluted by centrifugating and the system is washed. IgE antibodies are eluted after incubation of 10 minutes with 0.2M Glycine/HCl (pH 2.5), and immediately neutralized with 1M Tris (pH 9).

I have been doing some bibliogrpahic researh but the truth is that there is no much recent information regarding this. Does anyone recommend a specific protocol? Or anyone knows if I can send nmy samples somewhere for their purification?

Thanks!!
