To remove organics and carbonates from lake sediment samples for diatom enumeration, I was taught to boil sediment samples in 40ml concentrated nitric acid and then add potassium dichromate. After that I dilute with DI, let the suspension settle overnight, decant and then repeat the dilution/settling/decanting for a total of six times. This methods yield wonderfully clear and easy to count diatom slides. However, I'd like to not have to use the potassium dichromate. (And I wouldn't mind a speedier prep that avoids the several days of dilution/settling/decanting!) Suggestions?
