I am working on Drought stress tolerance on wheat, and it is found that polyamines play an important role in stress tolerance by modulating different physiological and biochemical process. i want to analyze polyamines content in plant tissues, (leaves, seed, and root). if anybody has protocol. how to measure polyamines content in plant tissues by spectrophotometer.
The analysis of poly amide by spectro photo metric method could be performed as follows:
1. 0.5-5 g of colored finished product is heated with 50 ml of distilled water and filtered.
2. It is acidified with acetic acid .
3. Add poly amide powder 0.5-1 g. and mix.
4. pour to chromatographic column.
5. Wash with hot distilled water followed by methanol. and discard washing.
6. collect the final wash of 5% methanolic ammonia.
7. Acidified the wash with acetic acid and evaporate it.
8. take absorption in spectro photometer using usual method.
9. The same sample of step 7 can be tested by TLC.

We can perform HPLC for the polyamines in additin to the spectrophotometric method.
