Dear Mustahsan,

Nothing is magic.

You have to test your homogenate with several dilutions. Until two dilutions are obtained, which will give identical results after calculation. You will then be in good working conditions. Measures at the same time the total protein content (bradford test for example), it will help you to optimize the dilutions of your different samples.

Thank you very much for your suggestion. I already measured this dilution factor last week. It needs around 180/200 time dilution for SD rat testis homogenate.
