I'm gonna to transfect iPSC derived neurons and have no idea which one to choose. Actually is the GFP-RFP-LC3 plasmid. My supervisor told me that if the promoter is CMV promoter, it will be silenced in the neurons. Does anyone has done similar experiments and have any experience about this?

FuGENE HD is for protein and virus production and delivery into primary and stem cells. It provides the highest efficiency compared to FuGENE 6.

I agree with your supervisor. Although CMV promoters are commonly used for high expression of transgenes in mammalian cells, they may be inappropriate promoters for strong expression in primary cells. Plasmids with the PGK1 or EF1α promoters express best in iPSC derived neurons.
