Does anyone have formulations for homogenization buffers for use with MAGPIX compatible:

1) protein multiplex panels

2) phosphoprotein multiplex panels.

I'm interested in solubilizing choriodecidua and amnion from fetal membranes. The amnion is particularly tough to completely solubilze even after multiple rounds of Tekmar homogenization, sonication and freeze/thaw.

For western blotting/2DIGE I've routinely used 2X-RIPA buffer with/without protease inhibitors, vanadate, and/or PMSF (which unfortunately interferes in Bradford-BIORAD protein assay). I have concerns that this buffer and/or these additives may interfere in the above multiplex assays.

I KNOW that most of the kit suppliers sell $$ Lysis Buffers containing their "proprietary" ingredients but, due to the large tissue samples I need to homogenize (must include particular representative areas) this would be costly.

Anyone have "tested recipies" (or links) for Homogenization Buffers compatible in the above assays that have defined "ingredients"?
