I have tried using the TBARS assay kit from caymanchem (10009055) and am not getting the changes I expect to see. For those who have used this kit, do you follow the protocol exactly as provided or are there some tips/tricks you can share? I've been using a sonicator as suggested in the protocol but it is quite ancient and I suspect it may not be doing the job. Do other lysis methods work just as well? Do the cells/tissues need to be fresh or will it work with a stored pellet/frozen tissue?
