I am curious to see if anyone has used this specific kit for samples containing single celled algae. When just collecting DNA we've found it useful to use enzymatic lysis with their Blood and Tissue kits. I've contacted Qiagen and they have said that our lysis methods are likely not compatible with this kit and that we should utilize beading if necessary.

The lysis Buffer RLT included in the AllPrep DNA/RNA/Protein Mini Kit is most likely not directly compatible with enzymatic pre-treatment procedures, although we have not specifically tested cellulase pre-treatment. The Buffer RLT contains a high concentration of guanidine salts, as indicated in Section 2 of the MSDS; the guanidine salts stabilize the RNA by inactivating RNases, however, other enzymes would also be expected to be inactivated. In addition, the Buffer RLT should not be significantly diluted, since the guanidine salts are important for binding of the nucleic acids to the spin columns.

Although it may be possible in principle to adapt the AllPrep DNA/RNA/Protein Mini Kit protocol to other sample types, such as microorganisms, we do not have a validated extraction procedure for algae. It may be possible to incorporate a mechanical lysis step, for example using a bead mill with glass beads in the undiluted Buffer RLT. This type of protocol modification would require validation by the user.
