I am hoping to recording brain oscillations from acute rat/mouse brain slices. I am looking for advice on how to proceed.
I want to record local field potentials in the GAMMA and Theta range. Recent reports show the amygdala has an intrinsic rhythmicity generator that can be modulated by endogenous and exogenous signals. I am hoping to record potential modulation of these oscillations.
You bath apply aCSF at physiologic temperatures and include aCSF in the recording electrode; I will have to adjust my high- and lowpass filters, according to experimental conditions.
I am using pClamp and multiclamp700B software. Can this all be done in Gap free?
Yes, you do that in gap-free mode.
When recording you do not need to set a high-pass filter.
Determine the highest frequency you want to be able to see (γ would go up to 80-100 Hz, but you may want to oversample by at least 10x to get some headroom for band filtering, post-processing etc, and end up with more detailed traces). Set a low-pass filter (on the amplifier - for antialiasing) right above that frequency, and then set your sampling rate (on the digitizer) to at least 2x the low-pass filter value.
Note that clampex has some high/low-pass filters next to the trace: these are not recorded in the file(!) but may be useful while recording.
