The PowerSoil® DNA Isolation Kit uses patented Inhibitor Removal Technology to isolate microbial genomic DNA from all soil types and environmental samples, as well as fecal, stool and biosolid samples. The isolated DNA has the highest level of purity, enabling successful PCR and qPCR amplification of organisms from the sample. The PowerSoil® DNA Isolation Kit distinguishes itself from MO BIO’s UltraClean® Soil DNA Isolation Kit with a humic substance/brown color removal procedure. This procedure is effective at removing PCR inhibitors from even the most difficult soil types. Environmental samples are added to a bead beating tube for rapid and thorough homogenization. Cell lysis occurs by mechanical and chemical methods.

Total genomic DNA is captured on a silica membrane in a spin column format. DNA is then washed and eluted from the membrane. DNA is then ready for PCR analysis and other downstream applications.

MO BIO offers a vacuum based protocol for faster processing without centrifugation for the DNA binding and column washing steps for Spin Filters. PowerSoil is too often presented as a perfect kit but it is not suitable for all type of samples and not for all soils. As any protocol, it has limitation. For example, PowerSoil was shown to do not be the most efficient protocol to extract DNA from acidic soil. Are your wine samples present a pH

I use a precellys 24 instrument  is better than Fast prep. with the PowerSoil kit that contains 0.7 mm crushed garnet with the bead tubes which can sometime break down a bit during the homogenization because of the power of the instrument.

Can anyone tell how much liquid sample requires for DNA extraction from power soil kit if we have to isolate DNA from liquid instead of soil?

The amount of water you need depends on how much cells you have in your solution, and how much sample you have access to. Usually, to extract DNA from liquid, you will filter your liquid and then extract DNA from the filter. You can use PowerWater from Mobio instead of PowerSoil:
