Single spore: seguencial dilution folowed by spread in agar water
For single spore isolates I use water agar and make dilutions from 10 to minus 2 to 10 to minus 6 and fish out the spores under reverse microscope.
Regarding morphological differences for ID the COONs agar gives very nice colony characteristics with addition of microscopy according to the spore characteristics should unable to differentiate Ascochyta pisi, A.pinodes and A.pinodella.
You can also grow the cultures of A.pinodella on Oatmeal agar for picnidium production and malt-extract agar to stimulate pigment production and crystal formation, and than perform NaOH spot test.
