I've tried amplifying a single band using matK primers for a while. The plant I'm dealing with also has many metabolites, including polyphenols. matK primers are also known to amplify mitochondrial DNA...it could be a reason. The GC% in the matK region is less than 50%. So, I would like to know if this is a common occurrence and if there are any methods to prevent double band formation.
