I need to get primary oocyte, secondary oocyte and ovary somatic cells from mature eggs in goldfish ovaries. I don't know how to separate them in mess-cells. I heard that they can be easily distinguished under a microscope. Then how would I digest the cellular junction? Does anyone know other methods?
The ovary cells of different types in fishes can be easily distinguished, but I never saw any paper dealing with the extraction of each of, or specifically the particular type of ovary cell from that organ. It is to detach mature eggs and extract them from other ovary cells manually and by bare eye, and then you would have other types of ovary cells in mass. However, the separation and extraction of other cell types need at least the use of a strong magnifying lens, if not the microscope. Than you can try to separate them using tiny dissecting tools, though I assume that would be too rough for your purpose and that you would need a kind of enzyme-involved procedure and size-related method to separate cells of different size and/or weight. If there is no yet a kind of ready-to-use method, than this is very interesting question.
