In TEM mode, with 5 kV, you have to make sections very very thin to pass through your sample, thinner than what you need for 80 kV TEM, but you don't have to contrast you sample because of this low voltage.
In SEM mode, I believe the sample is only restricted to one 3 mm diameter grid compared to traditional SEM with large chamber, in which you can have several samples of whatever sizes on whatever supports (big bugs, mammalian cells classical glass-coverslip etc.)
