I am going to inject a targeting mAb into xenograft mouse models (IP injection). Here are the questions:
1-How should I determine the dose of the antibody based on the in vitro work (this study is original)?
2-If I plan to inject two doses of the mAb, do I have to inject the exact equivalent doses of the isotype control as well?

2. Yes. Everything (the schedule, the dose, the buffer) has to be the same for your isotype control group.
While I do not have a specific answer, I think factors that you might want to consider include:
A-Potential half-life of the antibody
B-Nature and density of your target: What and how many cell types are expressing the target?
C-Objective of injection: Are you trying to deplete cells? Are you trying to block a receptor? Are you trying to trigger signaling (agonize) a receptor?
D-Nature of the antibody: Is there potential toxicity associated with injecting? Is the target critical for survival?
If you have enough of the antibody, and mice, I would suggest to start with 10 mg/kg (~250 μg/mouse) and titrate down.
