I am growing mouse bone marrow derived macrophages in cell culture.I am using MCSF or mouse-CSF from Peprotech.
The suggested ranges vary in the literature. I am using 100ng/ml. This costs a lot of cytokine. I do not wish to test dosage. I am isolating cells from mouse bone marrow that adhere to non TC treated plates and are treated with CSF.
Dose anybody have experience with recombinant CSF dose as far as selecting for adherent mouse bone marrow derived cells?

We use macrophage colony-stimulating factor at 20ng/ml. The lower level suggested by Nicole is probably just fine for short-term culture, but you may get better results if you have high density, longer-term cultures with a higher concentration of M-CSF.

I agree with Marcus that 20 ng/ml m-CSF is good for growing cells in couple days. Actually, I used this concentration m-CSF too.
Thank you for your answers. I am going to drop my current 100ng/ml dose by half and if that goes well I will be dropping further.
I hope this goes well, if so I have already "doubled" my cytokine (or halved my use thereof) and hope to save even more.

You could also try to use L929 conditioned medium, which is very cheap and works really well. Let me know if you want to give it a try and I will send you a protocol.
