Dear Hanyin, I am sorry for answering a different question but I consider it is important for the EEM issue. Did you think that some fluorophores have at least two excitation peaks and, by using the regional integration, the different excitation peaks may be counted, for a given fluorophore, by different integration areas? This point was discussed few times in the literature (and it is to me, quite obvious). That is why I am considering the FRI method as too empirical (and not sure it is much better that just regular peak picking). If to look for a better solution in the EEM analysis, I would recommend using PARAFAC. In this way, the spectral images obtained are not complicated by spreading excitation peaks over different integration areas. There is free software for PARAFAc And plenty people are using it now
