Hello everyone. I have done ELISA using coating antigens from different 'groups'. I am confused about whether the data of OD450nm values are normally distributed. Some of the researchers have used parametric tests like ordinary ANOVA and t-test depending on the number of comparing groups but I also found some papers in which the researchers have used non-parametric tests. Do not the non-parametric tests use for skewed data? I have done the analysis using different parametric and non-parametric tests and for some of my groups, the results/graphs are almost the same but for a few groups, there is a huge difference between the results/graphs/statistical significance. I am really confused about it.

Kindly help and guide me in this regard.
