We are currently producing AAVs in our lab, and have so far used the crude lysate to transduce our neurons in organotypic slice cultures. However, we'll now also need to purify and concentrate to get higher titers, as we need to transduce way more cells than we currently can with our crude lysate. We have tried purification with an iodixanol gradient and ultracentrifugation in the past, but would like to switch to something less time consuming that doesn't involve ultracentrifugation. I stumbled across AAV purification kits where you just run the lysate over a column, and thats it. Has anyone tried that, or has experience with that, and can tell me if this works nicely?
