To respective researchers, I’m doing the extraction of DNA from the tropical peat soil (high humic asid) for the metagenomics analysis. I’m using NucleoSpin Soil Extraction Kit (Macherey-Nagel). I got good in DNA yield but total failed in the purity. I want to know if anyone has used this kit? Or any recommendation for other kits or protocol?

Soil DNA extraction is usually complicated by the co- purified contaminants especially humic substances, which subsequently inhibit post - extraction analyses such as PCR- DGGE.. Various methods have been developed to remove humic substances from soil DNA, including the use of cesium chloride density cetrifugation, aluminium sulfate, hexadecyltrimethylammonium, polyvinylpolyrrolidone or PVPP, Sephadex G- 200 column. Mostly, it's due to humic property of peaty soil.... Try another DNA soil extraction kit. Mbio products is good for this ! Best of luck
