After I collect urine samples, I spin them down, some of these samples leave a large pellet that is pink in color. As I exctract these samples (using Qiagen RNeasy Kit) not only the precipitate get stuck on the column but always read with two peaks on the Nanodrop Denovix.
Why do you think this is happening, and whether you have experienced such a thing ?

But about your pinky pellet it's happened some times and it's because of oxalate if I am right, don't worry about that.

1. Centrifuge 5-15 ml of urine. In a 15 ml test tube. 2. Wash the precipitate with 1x PBS (buffer). 2-5 ml. We try to minimize the salt content. 3. Centrifuge again. 4. Dissolve the sediment in 100 µl 1x PBS. Transfer to a 1.5 ml tube. 5. Centrifuge at 14,000 rpm. 6. Remove the supernatant. And then you can add a buffer from the set to select immediately. Or add RNA stabilizer and store at -70 ° C.

In one of my research I extracted DNA from urine samples and those salts was not important to me but for RNA they can be important.
