I have read a paper in Nature on isolation of lpmc's from mouse colon tissue (2007, nb.10, page 2308). It is really interesting and I would like to give it a go. However, I am a bit concerned with the use of the digestive enzymes as they seem to contain LPS from the purification steps. I have not been able to locate these enzymes LPS free (except from DNAse). Have anyone had any problems with the cells being stimulated after isolation? I would like to carry out ex vivo experments on the cells after isolation. However, if cells are already stimulated from the enzyme solution I may need to reconsider. Maybe you could be so kind to tell me what your experience is with the stimulatory status of cells post isolation? Thanks
