Dear all,
Just wondering what is the difference between using PBS and MgCl2 for isolating micro-organisms? The specific protocol I'm looking at is this: https://www.nature.com/articles/s41596-020-00444-7 where they used MgCl2 to isolate and suspend bacterial cells for use in culturing; other protocols I've read also PBS or MgCl2.

PBS helps to maintain osmotic balance and it provides the cells with water and inorganic ions. Besides this, PBS also helps to maintain pH.
On the other hand, MgCl2 is used for the maintenance of the osmolarity of the solution.
So both PBS and MgCl2 serve the same purpose at the time of isolation and suspension of bacterial cells for use in culture.
