I wonder if anyone has seen the potency of inhibitors increase in the presence of detergent. I included 0.01% Triton in an assay which previously was not giving any hits and now I see a few! I have generally used this tactic to get rid of non-specific inhibitors/aggregators but now I am seeing the opposite effect. Any thoughts would be welcome.
The detergent may help keep hydrophobic inhibitors from sticking to the assay plate because the detergent coats the hydrophobic plate surface.
I will say yes. In fact, any added molecules in an assay may help (or not). I remember the negative effect of the addition of BSA on one polyphenol for example. Since the hydrotropic effect of ATP on the solubility of proteins (Science 2017), all the biomolecules may contribute to a biological response.
