Hi everyone. As part of an exposure, I need to feed C. elegans cultures deactivated E. coli OP50. We've had little success in deactivating the OP50 using freeze-thaw cycles and heat inactivation. Also, exposure in a UV box did not produce great results. The problem is that I either need to deactivate a washed OP50 pellet, or a suspension with an optical density of 3 (at 595 nm). So a very high concentration! Does anyone perhaps have any suggestions? Maybe a UV inactivation protocol (with details about exposure time, light intensity, what type of containers, etc.)? Or maybe autoclave at lower temp and pressure? Lysis should of course be avoided. Your help will be much appreciated! Thanks.
