I am looking for a calcium oscillation, conduction velocity and membrane potential analysis software or several different software that could analyze videos of cardiomyocyte stained with fluorescent dyes, particularly calcium dyes such as Fluo-4 or membrane potentials dyes such as Di-8-ANEPPS:

Average calcium peak:

  • Analyze single video file or sequence of image files and return single calcium peak, such as this: https://www.moleculardevices.com/sites/default/files/inline-images/examples-of-parameters-calculated-by-the-peak-pro-analysis-algorithm.png . I am particularly interested in a software that analyze fluorescence, since we are working with Cal 520 dye.
  • This is important since selecting multiple region of interest in a video file is time consuming. Moreover there are big variation between different region of interests. Selecting such regions manually might lead to data misinterpretation.
  • Since cells oscillate at different time periods, software should be able to align peaks and calculate the average.

Conduction velocity measurement:

I am also looking for a software for evaluating cardiac conduction velocity in iPSC derived cardiomyocytes. What dye should I use for such purposes? Maybe Di-8-ANEPPS? The software should be able to analyze single video file or sequence of image files and return how fast fluorescence travels. It could be some kind of vector motion software.

Membrane potential measurement:

Also I would be happy to hear any recommendations regarding the software for membrane potential measurement. Just as with calcium I am looking for a software that analyze single video file or sequence of image files and return single potential peak. Suggestions for single peak analysis software for membrane potential would be highly appreciated as well.
