I am setting up the HeLa cell xenograft model on Swiss nude mice, but I having trouble growing tumors. Has anyone tried growing tumors with this cell line? I tried without and with matrigel with a ratio of 1:1. The number of cells injected in both cases was 10 million cells subcutaneously. I don't get observing the tumor growth and would be glad if I could get any suggestions or help.
Do you harvest them during exponential growth phase? And are you avoiding trypsin when harvesting?

Yes, I injected cells when they were in the exponential growth phase. In relation to the second question, I use trypsin when harvesting. What do you recommend to me?

Depends on the experiment you want to do. I've been using NSG mice for xenografts or C57BL/6 mice for allografts.
Abhijeet Parkhe, trypsin will digest cell-surface proteins that may be necessary in the early steps of xenograft formation.
Concentration, incubation time, cell type etc.
Trypsin is widely used. Hence both the pros and cons are well known. Trypsin is usually an issue when the readout is about cell-cell and cell-matrix adhesion.
