I have 5 thermophilic bacteria that had been preserved for a year in glycerol solution (15%) at -80 oC. I tried to grow the five isolates in LB broth, 50 oC, 150 rpm, but only 3 isolates grew after 24 and 48 hours. The rest (2 isolates) did not grow until tenth day of incubation. Are the two thermophilic isolates loss their ability to grow at high temperature because of the preservation?
Delayed growth after deep freeze is a common phenomenon. They did grow in 50C, so no worries...
Their ability to grow at high temperature should not change, that would be genetically defined. However the survival of those cells at -80 might be variable. But once you get them to grow they should be identical to the strains you stored.
As already mentioned by Dr. Benedik, the survival of the cells at -80 might be variable and depends on the methodology followed. Sometimes, the methodology followed for cryopreservation effect the revival rate of bacterial cells. First revive the isolates and then check the thermophilic growth characteristics.
