I have 5 thermophilic bacteria that had been preserved for a year in glycerol solution (15%) at -80 oC. I tried to grow the five isolates in LB broth, 50 oC, 150 rpm, but only 3 isolates grew after 24 and 48 hours. The rest (2 isolates) did not grow until tenth day of incubation. Are the two thermophilic isolates loss their ability to grow at high temperature because of the preservation?
