I am working with a plant calmodulin-binding protein (CaMBP). The calmodulin (CaM) is very important protein in Ca2+ signaling. In order to know the relationship of my CaMBP and calcium, embryogenic calli were cultured in MS liquid medium supplemented with different concentration of CaCl2 (0, 1, 2, 3, 4, 5, 6 mM) for 24 h. After running QPCR, the relative expression level of my interested gene (CaMBP) is significantly much lower in MS medium with CaCl2 than that of in MS without CaCl2. (I repeated that experiment. But the result was the same as previous.). I don't know the reason why the expression level was low in medium with CaCl2. I hope that someone can kindly explain that phenomena. Thank you in advance.
