For example, if a tryptic peptide (spanning its N-terminal) could be detected in PMF, and in MS/MS it also matched with a reasonably high Mascot ion score.

With such information, do I have sufficient evidence to say that I have 'confirmed' the N-terminal sequence of the cloned protein? or Would I need an extra 'Edman N-terminal sequencing confirmation'?

What is the common way researchers use for such confirmation nowadays?
