Though I know 5% phenol solution means 5 g phenol crystal for 100 mL distilled water, but, I am concerned about melting of phenolic crystal in water using heat. Could anyone suggest me how can I do that?
Dear Sir. Concerning your issue about the preparation of 5% phenol. I think the following below links may help you in your analysis:
https://stockingerlab.osu.edu/sites/stockinger/files/imce/PDFs/Protocols/PhenolPrep.pdf https://link.springer.com/content/pdf/bbm%3A978-94-010-9139-8%2F1.pdf

Thanks
Dear Abdul,
You may use certain buffers such as TrisCl or TE buffer.
You may adopt your procedure from here: http://www.mrc-lmb.cam.ac.uk/ms/methods/phenol.html

Phenol is moderately soluble in water - about 8 g of phenol will dissolve in 100 mL water. So, take 5 gm phenol crystals and add water to 100 mL.
Here,
this might help.
