Pleae try to use "Trypan blue". Trypan blue is a generally stain for select dead cell or living cells. Live cells are not colored, but if cell dead will turn to blue color.
3-[4, 5-dimethyl-2- thiazolyl]-2, 5-diphenyl–2H tetrazolium bromide (MTT), this chemical is very useful for lympho proliferative assays. You can see more about it in an artiicle titled Chemotherapy of Rhinosporidiosis: a Review by S N Arseculeratne
I used 1 ug/ml propidium iodide and FACS. It worked pretty well for L. infantum. There is no reason why not working for L. amanzonensis. Give it a try. Good luck.
Trypan blue solution (0.2%) in saline solution containing 0.01% sodium aside, works well and after 2 minutes, you can check the Leishmania promastigotes, we have used it with different Leishmania species.
I tried to use trypan blue (0.4 % in normal saline) to differentiate dead and live L.major, but the dead leishmania that was circle didn't absorb the dye, then it wasn't work perfectly. Had anybody the common experience. thank you for your comments
