Conjugation of amine modified DNA with iron oxide nanoparticle ?

Hi everyone,

I plan to conjugate DNA with iron oxide NP via the amine-carboxyl linker. I wonder if someone can comment on my experiment conditions:

+ pH: TE buffer, pH~7

+ reaction time: 30 mins for EDC/NHS activation; overnight for DNA-iron oxide conjugation

+ EDC/iron oxide molar ratio: 15

+ DNA/iron oxide molar ratio: 0.5 (I want to get 1 DNA per particle)

I think that several points are problematic in your protocol. You should search for a similar experiment in the literature and try reproduce it.

The first problem is your buffer. Tris is absolutely incompatible with EDC/NHS chemistry.
