You should certainly use fixed excitation and emission slit and PMT voltage.
ideally, you should use the same excitation wavelength, and either fixed concentration, or vary the concentrations so that the absorbance at the excitation wavelength is the same for all.
I fully agree Dr. Hugh J Byrne , one more addition, the concentration must be adjust low enough to avoid re-absorption
I measure the FL of compounds in benzene, do I have to measure the FL of benzene only then subtract the FL of the benzene of FL of the sample?
