I have recently inquired information about synthesis of magnetic nanoparticles coating a peptide. Although there were many articles related to this issue but I cannot find any information about the amount of coating agent, the concentration of it as well as the drop falling rate. So who has experiences in this field, please share with me? Thank you a lot!

Apart from the surface charge compatibility and the need to cross linker which are very important issues, many routes are there to find the suitable amount of molecules to be loaded on the surface of nanoparticles. However, the following steps are essential:
1- Prepare a dilute suspension of the nanoparticles, like 1 g/L, in a suitable media, aqueous or/and non-aqueous with or without additives, and treat the suspension with ultrasonic for better dispersion.
2- Dissolve the peptide in a suitable solvent. It is better to use, if possible, the same media of the suspension as solvent. The best concentration, which is enough to totally cover the surface of the nanoparticles without excess amount, can be roughly estimated by dividing the measured surface area of the nanoparticles by the average molecular projection area of the peptide molecule. However, the best concentration can be found experimentally by adding known amount of peptide to the suspension of the nanoparticles, under vigorously stirring, and leaving the mixture to equilibrate for 1 h at room temperature followed by centrifuging to separate the coated nanoparticles. Quantify the amount of peptide left in solution, by any available method, to find the amount consumed peptide for coating.
3- Add the peptide solution to the suspension, or vice versa, drop by drop with vigorously stirring and sonication.
