I am currently working on the determination of enzyme activity by using Polyphenol Oxidase(PPO) Assay Method. In this method, i am using polyvinylpolpyrrolidone(PVPP) mixed with citrate buffer solution and catechol solution to obtain data using the UV-Vis Spectrophotometer. The current condition of my catechol solids is that it turned into brown colour and smelly (most probably oxidised already). The question is, is it still useable in that condition for my methods?
From my experience I would suggest to discard your catechol. It might not be usable anymore. Even though some catechol molecules might not be polymerized yet, the background level for your photometric assay might be too high (/bad). I am wondering that you catechol is of so unstable nature. I would suggest to use the next time gaseous nitrogen to flush you bottel carefully after using.
